I bought an aluminum enclosure for a 2.5" HDD, but I have questions. The seller doesn't know how to answer them, and there's no manual:
1) Is there a risk that the HDD PCB will touch the aluminum and cause a short circuit?
2) I received a sponge in the package, and it has a possible division in the center. What is the use of this sponge? Where should I install it correctly? Will the HDD have physical problems without this sponge?

Yes. You may use some kind of insulation tape to separate the case from the disk drive PCB.

Reduce noise/vibration by filling the gap between the disk drive and the case, so this will ensure that the drive should remain fixed in the case.
It is better to use it.
